The3C-likeprotease(3CLpro)ofsevereacuterespiratorysyndromeassociatedcoronavirus(SARS-CoV)isvitalforSARS-CoVreplicationandisapromisingdrugtarget.Recombinant3CLprowasexpressedinPichiapastorisGS115asa42kDaproteinthatdisplayedaKmof152withDabcyl-KTSAVLQSGFRKME-Edansassubstrate.Purified3CLprowasusedforinhibitionandkineticassayswithsevenflavonoidcompounds.TheIC50ofsixflavonoidcompoundswere4781.Quercetin,epigallocatechingallateandgallocatechingallate(GCG)displayedgoodinhibitiontoward3CLprowithIC50valuesof73,73and47,respectively.GCGshowedacompetitiveinhibitionpatternwithKivalueof251.7.Inmoleculardockingexperiments,GCGdisplayedabindingenergyof?14kcalmol?1totheactivesiteof3CLproandthegalloylmoietyat3-OHpositionwasrequiredfor3CLproinhibitionactivity.
